The researchers say the discovery could lead to the development of new drugs for cancer treatment.
Fluorescent proteins are used as an excellent marker in molecular biology, so knowing how to change one is important. This new study, published this week in the journal Science, shows that protein engineering can be used to turn a fluorophore, or green fluorescent protein, into a red one by swapping out some of its protein building blocks.
“We found that by making small changes to the sequences, we can achieve a completely new function,” said Jianfang Feng, co-author of the study from the Institute of Zoology in the Chinese Academy of Sciences. “The most important accomplishment in our paper is that we can force these new functions to appear from non-functional proteins.”
Previous research has used evolution and protein engineering to build proteins that could turn themselves on by having the amino acid tryptophan change its fluorescence. The ability to make such a change has allowed scientists to make a protein that detects a specific type of cell.
The new research demonstrates how to change a protein’s fluorescence from green to red. So far, using this technique, the team has only been able to turn fluorescence into lethal activity. The next step will be to find out how to turn the fluorescence back on to see if they can apply the technique to other applications as well.
