Guitar Hero: On Tour is set in a fictional universe of rock stars and performing groups, and the game follows their journey for a music concert. The story begins with the formation of the original band, which consists of five members, who work to earn enough money to buy a van to take to the Iron City Rocks concert.[63] The game follows the rise of the band members, their success, and their break-up, all of which are traced over five decades of years, ranging from 1958 to 2008.[63]

The game follows three points of view: the guitarist, who continuously plays guitar and sings while the girl’s songs are played by the band members; the lead singer, who is the only member of the original band who manages to stay on the band, trying to make the famous boy’s band career to succeed and finally leave the band; and the drummer, who looks after the opening and closing of the concerts and records the band’s songs. In the court of Metalville, members of the band compete to become “Iron City Rocks singer” (a fake rock star). The Guitar Hero versions are slightly different in the lead singer, being divided into two characters, and the guitarist playing their own version using the Nintendo DS hardware’s microphone and buttons.

In the game, either the guitarist or drummer can use any number of instruments to perform the song. For example, the guitarist plays guitar, while the drummer plays drums, or the drummer plays drums with the bass guitar, or the bass guitarist plays bass with the guitar.
